چکیده

DNA complex has been widely used as non-viral vectors for the delivery of genes or siRNA. Owing to the strong and long-ranged electrostatic interaction, the structure and property of the DNA complex should evolve with time in a long-term. To test this hypothesis, we choose 2000 bp double-stranded DNA and 21 bp oligonucleotide as the model molecules, and comparatively studied their complexation with narrowly-distributed poly-L-lysine (PLL150) by time-resolved laser light scattering. In the time range of about one week, the complexation of both DNA samples underwent three stages: formation of preliminary complex, further aggregation, followed by precipitation. The aggregation and precipitation rate of the complex formed by oligonucleotide was much faster than that of the complex formed by 2000 bp dsDNA. After precipitation, the amount of the longer chain polyelectrolyte, as determined by UV and fluorescence, was about twice that of the short chain polyelectrolyte in the sediment. The precipitates were far from being fully neutralized. Moreover, the component ratio in the sediment was independent of the mixing charge molar ratio. A rational complex mechanism was proposed on the basis of these findings. During complexation, the relaxation of polyelectrolyte inside the complex and the exchange of polyelectrolyte between complex determined the aggregation and precipitation rate. The competition of the two kinetic processes governed the structure and property of the complex in the solution and in the

Inhibition of nonenzymatic depurination of nucleic acids by polycations

DNA base depurination is one of the most common forms of DNA damage in vivo and in vitro, and the suppression of depurination is very important for versatile applications of DNA in biotechnology and medicine. In this work, it was shown that the polycations chitosan (Cho) and spermine (Spm) strongly inhibit DNA depurination through the formation of polyion complexes with DNA molecules. The intra...

سالیتون‌های توپولوژیک در مولکول DNA با پتانسیل اصلاح یافته

DNA is not only an essential research subject for biologists, but also it raises very interesting questions for physicists. The open states in DNA double helix can lead to topological solitons. Since DNA is a very long molecule )of order a meter or so long) and nano-scale width, solitons can propagate along the molecule. In this paper, considering a correction term in the interaction potential b...

Studies on electron beam induced DNA damage and repair kinetics in lymphocytes by alkaline comet assay

Background: Exposure to ionizing radiation is known to induce oxidative stress followed by damage to critical biomolecules like lipids, proteins and DNA through radiolysis of cellular water. Since radiation has been widely used as an important tool in therapy of cancer, the detailed investigation regarding the DNA damage and repair kinetics would help to predict the radiation sensitivity of cel...

Transferrin-polycation-DNA complexes: the effect of polycations on the structure of the complex and DNA delivery to cells.

We have previously described a gene delivery system based upon the receptor-mediated endocytosis of DNA complexed with transferrin-polycation conjugates. This delivery system has been found to be very effective for both the internalization and the expression of genetic material in cells that have many transferrin receptors. Upon scrutinization of the parameters involved in this method, which we...

Bio-reducible polycations from ring-opening polymerization as potential gene delivery vehicles.

Synthetic polycations show great potential for the construction of ideal non-viral gene delivery systems. Several cationic polymers were synthesized by the epoxide ring-opening polymerization between diepoxide and various polyamines. Disulfide bonds were introduced to afford the polymers bio-reducibility, while the oxygen-rich structure might enhance the serum tolerance and biocompatibility. Th...
